The W3C-WAI (Web Accessibility Initiative) guide lines for Accessibility explained. This is the gold-standard group that posts and makes the standards for the internet. We use their specifications and Validator, as well as their guide lines and examples on how to make this site fully Accessible. 
There are a number of other commercial and non-commercial validating concepts and services available.
The Bobby Validator will check for USA 508 Guideline compliance. It will show your page in the reader screen and then tell you if it passes or not, It also tells you what needs to be checked manually also as the online tester is not set up for every kind of text.
